Eli Lilly vs. Novo Nordisk: Has GLP-1 Manufacturing Become the New Competitive Moat? 

The obesity-drug market has largely been viewed as a race between clinical innovation and commercial execution. Investors have spent the past three years comparing prescription growth, market share, and pipeline candidates as Eli Lilly (NYSE:LLY) and Novo Nordisk (NYSE:NVO) compete for dominance in one of healthcare’s fastest-growing markets.

Yet another competitive advantage for investors to keep in mind is manufacturing.

During the early stages of the GLP-1 boom, demand consistently exceeded supply. Shortages of Wegovy, Ozempic, Mounjaro, and Zepbound demonstrated that developing a breakthrough medicine is only part of the challenge. Companies must also manufacture those medicines reliably, expand production fast enough to meet demand, and build enough capacity to support the next generation of therapies.

The company has invested heavily in expanding its Clayton, North Carolina campus while strengthening its fill-finish capabilities through the acquisition of former Catalent facilities. Since three decades, the company has expanded nine times in North Carolina, invested nearly $6 billion dollars, built and upgraded two more facilities, and expanded its employee count from 50 to more than 2,000.

In addition to expanding its U.S. footprint, the company’s manufacturing network remains more geographically diversified than Lilly’s (NYSE:LLY), which has historically been an advantage from an operational perspective. This approach also emphasizes manufacturing experience rather than manufacturing scale alone. Producing complex biologic medicines consistently and efficiently requires years of operational knowledge, and Novo remains one of the world’s largest manufacturers of insulin and GLP-1 therapies.

Bear Case

The biggest risk for Lilly (NYSE:LLY) is that manufacturing capacity eventually becomes abundant rather than scarce. Extraordinary demand for obesity medicines is one of the factors driving increased investment in its operations, but the competitive landscape is evolving rapidly. Novo Nordisk (NYSE:NVO) continues expanding production, while several other pharmaceutical companies are developing competing obesity therapies. If supply eventually catches up with demand, manufacturing capacity may become less of a competitive differentiator and more of a fixed cost. That would place greater pressure on Lilly (NYSE:LLY) to generate attractive returns from one of the largest manufacturing expansion programs in the pharmaceutical industry.

Novo Nordisk (NYSE:NVO) faces a different challenge. While the company possesses extensive manufacturing expertise, it has also experienced periods where demand outpaced available supply. Those shortages suggested that operational excellence alone cannot overcome insufficient production capacity. As the obesity market continues expanding, Novo Nordisk (NYSE:NVO) should continue expanding its manufacturing capacity quickly enough to support future prescription growth without supply constraints.

There is also a strategic question surrounding manufacturing flexibility. Lilly’s (NYSE:LLY) broader investments across multiple production technologies may ultimately provide greater optionality if obesity treatments evolve toward new drug modalities or if future pipeline assets require different manufacturing processes. Novo’s (NYSE:NVO) existing manufacturing network could prove highly efficient for current products, and could work in the future as well if it remains adaptable to the industry’s manufacturing requirements that may change significantly over the next decade.

Which Manufacturing Strategy Is Better?

The comparison ultimately reflects two different philosophies. Lilly (NYSE:LLY) is investing aggressively today to build manufacturing capacity that can support both current demand and future innovation. The strategy requires substantial capital but could create a durable competitive advantage if demand remains strong and the company’s pipeline continues delivering new products.

Novo Nordisk (NYSE:NVO) is relying on decades of manufacturing expertise while expanding capacity where it sees the greatest need. That approach requires less capital and may ultimately produce higher returns if future demand grows more gradually than many investors expect.

Neither approach is inherently superior. Their success will ultimately depend on whether future demand justifies Lilly’s (NYSE:LLY)  aggressive investment or rewards Novo’s (NYSE:NVO) more measured expansion strategy.
